Scope and Application The method is applicable for confirming presence of β-Lactam (ampicillin, nafcillin, cefazolin, desfuroylceftiofur cysteine disulfide metabolite of ceftiofur, penicillin G and desacetyl cephapirin) residues in bovine and porcine kidney and muscle. This method may also be used to screen for amoxicillin, cloxacillin, dicloxacillin and oxacillin in the same matrices.
Method Summary The β-Lactams are extracted from thawed lean tissues by homogenization and centrifugation using acetonitrile/water followed by clean-up using solid phase extraction. Penicillin V is used as an internal standard The final eluate is examined for presence of β-Lactams by LC/MS/MS using a triple quadrupole mass spectrometer under electrospray ionization conditions. A C18 waters, YMC ODS-AQ column is used for separation of the analytes.
Applicable Concentration Range The analytes may be detected in bovine and porcine kidney and muscle at the following concentration levels: ampicillin ≥ 10 µg kg-1; nafcillin ≥ 20 µg kg-1; cefazolin ≥ 50 µg kg-1, desfuroylceftiofur cysteine disulfide metabolite of ceftiofur ≥ 50 µg kg-1, penicillin G each ≥ 50 µg kg-1; and desacetyl cephapirin ≥ 100 µg kg-1. Screening of amoxicillin and cloxacillin at ≥10 µg kg-1 as well as dicloxacillin and oxacillin at ≥ 50 µg kg-1 is also possible.
