| Method Summary |
Ground muscle tissue carefully trimmed to avoid fat and connective tissue is blended and extracted repeatedly in ethyl acetate by centrifugation. The bulk of ethyl acetate is evaporated before adding aqueous sodium chloride to the extract. The remaining ethyl acetate is completely evaporated and the mixture then centrifuged after adding hexane to remove fat. The lower aqueaous layer is passed through a C18 cleanup column from which Chloramphenicol is eluted using acetonitrile. The eluate is then evaporated to dryness before derivatization at 65 ± 5 °C in a sand bath or water bath for 15 min using Sylon HTP. This derivatization agent is then completely evaporated and the residue reconstituted in cyclohexane/hexane (60:40, v/v) prior to analysis by GC/MS (quadrupole mass selective detector). Capillary inlet, splitless injection fitted crosslink methyl silicone column is used. |

Chloramphenicol may be detected, confirmed and quntified in bovine and turkey muscle at the concentration levels ≥ 0.25 µg kg-1 and ≥ 0.3 µg kg-1, respectively. |
