The method is applicable for screening/confirming the presence of aminoglycosides (amikacin, apramycin, dihydrostreptomycin, gentamicin, hygromycin B, kanamycin, neomycin B, spectinomycin, and streptomycin) in bovine kidney, liver and muscle. |
| Method Summary |
The aminoglycoside residues are extracted by centrifugation from bovine liver, kidney and muscle tissue (free from fat and connective tissue) using a buffer containing ammonium acetate/trichloroacetic acid (NH4OAc/EDTA/NaCl/TCA buffer). Tobramycin is used as an internal standard. The sample extracts (adjusted to pH 6.50 ± 0.05) are cleaned-up with a weak-cation exchange sorbent using disposable pipette extraction followed by elution with 10% formic acid in water. The final extract is analyzed using ion-pair reverse phase UHPLC/MS/MS in positive electrospray ionization mode. A C18 waters UPLC BEH column is used for separation. |

Amikacin, apramycin, dihydrostreptomycin, gentamicin, hygromycin B, kanamycin, neomycin B, spectinomycin may be detected in bovine kidney, muscle, and liver at various levels as detailed in the attached SOP. |
