Scope and Application The method is suitable for the identification of Chlortetracycline hydrochloride, Oxytetracycline hydrochloride and Tetracycline hydrochloride in bovine, porcine, ovine muscle, kidney and liver as well as poultry muscle and kidney.
Method Summary Fresh or partially thawed samples carefully dissected to avoid fat and connective are finely minced or ground and extracted with Mcllvaine buffer/EDTA solution by centrifugation. The extracts are then filtered through a GFB filter paper and cleaned-up by passing through C18 SPE catridges. The tetracyclines are eluted from the cartridge using methanolic oxalic acid, evaporated and reconstituted with aqueous methanol prior to analysis using a DIONEX Ultimate 3000 UHPLC connected to a VWD-3400 detector. A Syncronis C8 analytical column was used for separation.
Applicable Concentration Range Using this method, Chlortetracycline hydrochloride, Oxytetracycline hydrochloride and Tetracycline hydrochloride may be quantified at ≥ 500 µg kg -1 concentration level in bovine, porcine, ovine muscle, kidney and liver as well as poultry muscle and kidney
