The method is suitable for the identification of Chlortetracycline hydrochloride, Oxytetracycline hydrochloride and Tetracycline hydrochloride in bovine, porcine, ovine muscle, kidney and liver as well as poultry muscle and kidney. |
| Method Summary |
Fresh or partially thawed samples carefully dissected to avoid fat and connective are finely minced or ground and extracted with Mcllvaine buffer/EDTA solution by centrifugation. The extracts are then filtered through a GFB filter paper and cleaned-up by passing through C18 SPE catridges. The tetracyclines are eluted from the cartridge using methanolic oxalic acid, evaporated and reconstituted with aqueous methanol prior to analysis using a DIONEX Ultimate 3000 UHPLC connected to a VWD-3400 detector. A Syncronis C8 analytical column was used for separation. |
