Scope and Application This method is suitable for analysis of the Sulphadiazine, Sulphathiazole, Sulphapyridine, Sulphamerazine, Sulphamethazine, Sulphamethizole, Sulphamethoxypyridazine residues in meat sample.
Method Summary Mince tissue mixed with HCl, Na2SO4 (except chicken muscle) and ethyl acetate is extracted by centrifugation after homogenization with an Ultra-Turrax. Aliquots of the supernatant are evaporated to dryness and the residue dissolved in methanol, glacial acetic acid, water and hexane prior to further centrifugation. The aqueous layer is then analysed by HPLC-UV-Vis following post-column derivation using p-dimethylaminobenzaldehyde (after eluting the sulfonamides from the analytical column, the analytes are passed through a PTFE reaction coil where derivatisation occurs). The deriavatized analytes are detected at 450 nm. A Phenomenex C-18 Luna analytical column is used for separation.
Applicable Concentration Range The residues of Sulphadiazine, Sulphathiazole, Sulphapyridine, Sulphamerazine, Sulphamethazine, Sulphamethizole and Sulphamethoxypyridazine may be detected and quantified in chicken meat and porcine kidney samples at the concentration level ≥ 50 µg kg -1.
