Scope and Application Up to 56 multiclass pesticide residues can be screened in ovine, caprine, poultry, porcine, and bovine muscle by LC/MS/MS and GC/MS/MS.
Method Summary Various classes of pesticides are extracted from muscle tissue (20 grams plus or minus 0.2) with high speed dispersion in ethyl acetate followed by solvent exchange to acetonitrile and clean-up using ultra-low temperature freezing, centrifugation, and solid phase extraction following a QuEChERS principle. After further cleanup with primary secondary amine and MgSO4, extracts are pressed through a 0.2 micrometer filter before LC/MS/MS analysis. GC/MS/MS samples are eluted in acetone/toluene and after most of the solvents are evaporated, the injectable portion is dissolved in toluene. Analytes are separated on Bruker VF (5ms 30 m, 0.25 mm) and Waters Acquity UPLC HSS, T3 (1.8 micrometer, 2.1 by 100 mm) columns, respectively.
