Scope and Application The method is suitable for screening the presence of nitrofuran metabolites 3-amino-2-oxazolidinone (AOZ) and 3-amino-5-morpholinomethyl-2- oxazolidinone (AMOZ) in liver of food animals.
Method Summary Blended liver samples in deionized water, HCl, and 2 nitrobenzaldehyde in DMSO are vortexed and incubated for 16 hours at 35 to 39 degrees celcius prior to mixing with K2HPO4, NaOH, and ethyl acetate. The mixture is then extracted by centrifugation before evaporating the mixture to dryness. The residue is redissolved in n hexane/heptane and mixed with a sample buffer prior to re-centrifugation. The organic layer is then discarded and the remaining buffer containing AOZ and AMOZ derivatives analysed by ELISA. Absorbance is measured at 450 nm against an air blank within 60 minutes of addition of stop solution.
Applicable Concentration Range AOZ may be detected in bovine, porcine and avian (poultry) liver at ≥ 0.5 µg kg -1 concentration level. While AMOZ may be detected at the same concentration (≥ 0.5 µg kg -1) in bovine and porcine liver, its detection limit in avian liver is twice as high (≥ 1 µg kg -1).
